Class: Human Milk Oligosaccharide (HMO)
Format: Powder
MyOli® 6′-SL (6′-Sialyllactose)

Humans are unique in that at birth our brains are only 25% of adult size. Sialic acid plays an essential role as a building block for the brain during early life’s rapid development. Sialylated HMOs, such as 6'-SL in mothers’ milk, represent the main biological source for sialic acid. It’s found in high concentrations in human milk during the initial days of life.
Recent research also points to positive effects on the immune system, where it has been tested in clinical trials of adults1, 2.
ABOUT HMOs: The third largest component of mothers’ milk solids, HMOs are unlike regular sugars and don’t provide energy to the infant. Rather, they are prebiotics that feed an infant’s microbiota. They are also bioactive and deliver direct health benefits on their own. Each mother’s milk has a unique combination of HMOs, dependent on genetics, nutrition, lactational stage, mother’s lifestyle, geography and more3.

Health benefits
GUARDS AGAINST UNWANTED MICROBES
HMOs have multiple methods to rid the body of unwanted microorganisms, such as acting as decoys or altering epithelial cell gene expression4, 5. Each HMO has a highly specific mode of action6.
6'-SL can act as a decoy to attract certain unwanted microbes to attach to it instead of the epithelial surface, thereby ensuring they are flushed out of the system without causing harm7, 8, 9.
ESTABLISHES AN OPTIMAL GUT MICROBIOME
HMOs are prebiotics that promote beneficial bacteria growth4, 10. Long known to attract the healthy Bifidobacteria, recent studies highlight that over 100 non-Bifidobacterium species also use HMOs for their growth11. These bacteria then thrive and produce the metabolites necessary to support good health12.
Like other sialylated HMOs, 6'-SL only promotes moderate growth of some bacteria like infant-type Bifidobacterium infantis and B. bifidum13, 14. But it may help maintain stability of the gut microbiota under stress conditions and may exert effects on stress-related behavioral outcomes through modulation of the microbiota-gut-brain axis15.
PROMOTES A HEALTHY GUT
HMOs may promote mucus production16, thereby supporting the physical barrier between the intestinal epithelium and gut content. As well, metabolites produced by HMO fermentation, like short-chain fatty acids, can modulate the immune system and strengthen the gut barrier17.
6'-SL has been shown to promote epithelial cell differentiation and maturation, which may contribute to improved intestinal barrier integrity18, 19.
MATURES THE IMMUNE SYSTEM
HMOs interact with immune cells both locally in the intestine and systemically, supporting immune cell balance and immune signaling pathways. Their effects are mediated both via direct interactions with immune cells and via metabolites generated during HMO fermentation.
Like 3'-SL, 6'-SL helps shape immune responses, for example, by promoting TH1 and TH17 responses. These stimulate the immune system to fight. Both 6'-SL and 3'-SL can also reduce TH2 response, which is associated with skin discomfort20.
SUPPORTS BRAIN DEVELOPMENT AND COGNITION
HMOs are a source of sialic acid and fucose, both of which are essential components for brain development. They also play key roles in neurological processes. A small proportion of certain HMOs can enter the bloodstream, where they may exert positive systemic effects. In addition, HMOs are fermented into microbial metabolites that influence the microbiota–gut–brain axis.
6'-SL is, like other sialylated HMOs, a key a source of sialic acid, an essential building block for the brain. It’s also positively associated with cognitive development and motor scores21. Beyond that, studies link 6'-SL with supportive effects on brain structure and cognitive outcomes22.
PERFORMANCE IN ADULTS
6'-SL has been evaluated in clinical trials in adult populations, where its safety and tolerability were consistently confirmed, including at daily intakes of up to 6 g2.
Beyond safety, clinical research has shown that 6'-SL can enter the systemic circulation. In addition, supplementation has been associated with improvements in proximal muscle power2 and a slowing of muscle strength decline1.
See what science tells us
HMOs represent the beginning of complex processes where it's difficult to determine cause and effect. Much more research is needed to clarify their individual and collective benefits. Here’s a summary of what we know now.
MyOli® 6'-SL is part of our MyOli® 5 HMO Mix, which is custom blended for your specific target market.

More about HMO science
Breastmilk is the gold standard in infant nutrition. A major difference between milk from humans and other mammals is the amount and abundance of oligosaccharides. Back in 2015, when we were the first to make HMOs commercially available with our MyOli® brand, HMOs were mainly known for their prebiotic benefit.

Breast milk nourishes baby’s microbiome
Mother’s milk has evolved to give a baby and its microbiome a good start in life. Human milk oligosaccharides can’t be digested by the body— only by bacteria in microbiome.
HMOs are the third largest solid component.

Over 200 HMOs belong mainly to three classes
The vast majority of HMOs belong to one of three classes. 70% contain fucose and are called Fucosylated HMOs. Sialylated and neutral HMOs split the remaining 30%. SL HMOs contain sialic acid. Neutral HMOs have neither fucose or sialic acid, and are building blocks for more complex molecules4.

Three HMO classes with many health benefits
Each HMO is unique and offers multiple benefits. In general, FL HMOs tend to be good at protecting against unwanted microbes. The sialic acid in SL HMOs is important to brain and cognitive function. Neutral HMOs nurture a healthy microbiome23. This is just the tip of the iceberg for HMO health benefits.

Leading the science
As much as is known about HMO health benefits, more is waiting to be discovered. Some of our own scientists and collaborative partners are world-renowned for their HMO research. We also have a donation program so independent researchers can conduct their own studies.
