Class: Human Milk Oligosaccharide (HMO)
Format: Powder
MyOli® 2'-FL (2′-Fucosyllactose)

2'-FL has been studied more than any other HMO, and with positive outcomes for people of all ages. Research shows that it provides a variety of health benefits ranging from acting as a prebiotic1to supporting healthy cholesterol2. In fact, it has the broadest range of health benefits of any HMO, including language development and brain formation3,4.
While concentrations vary widely, 2'-FL is the most abundant HMO in mother’s milk and can easily represent 32% of the overall HMO amount5. As with most HMOs, 2'-FL decreases over time during lactation, but it typically remains the predominant HMO in breast milk6.
ABOUT HMOs: The third largest component of mothers’ milk solids, HMOs are unlike regular sugars and don’t provide energy to the infant. Rather, they are prebiotics that feed an infant’s microbiota. They are also bioactive and deliver direct health benefits on their own. Every mother’s milk has a unique combination of HMOs, dependent on genetics, nutrition, lactational stage, mother’s lifestyle, geography and more7.

Health benefits
GUARDS AGAINST UNWANTED MICROBES
HMOs have multiple methods to rid the body of unwanted microorganisms, such as acting as decoys or altering epithelial cell gene expression8, 9. Each HMO has a highly specific mode of action10.
2'-FL acts as a soluble receptor decoy that mimics the structures of epithelial cell surfaces. This attracts unwanted microbes and reduces their attachment to intestinal and respiratory epithelial cells. Based on this mechanism, emerging science indicates 2’-FL may contribute to promoting gut comfort11.
ESTABLISHES AN OPTIMAL GUT MICROBIOME
HMOs are prebiotics that promote beneficial bacteria growth9. Long known to attract the healthy Bifidobacteria, recent studies highlight that over 100 non-Bifidobacterium species also use HMOs for their growth. These bacteria then thrive and produce the metabolites necessary to support good health12.
2'-FL is a key probiotic nutrient for the infant microbiome; it selectively attracts bacteria beneficial to the early-life microbiome, including many Bifidobacterium Infantis and Bifidobacterium Bifidum species. It can also be utilized by other health promoting bacteria such as Akkermansia muciniphila and Bacteroides species13, 14, 15. As a result, 2'-FL may help shift the gut microbial composition toward the pattern typically seen in breastfed infants16, 17.
PROMOTES A HEALTHY GUT
HMOs may promote mucus production18, thereby supporting the physical barrier between the intestinal epithelium and gut content. As well, metabolites produced by HMO fermentation, like short-chain fatty acids, can modulate the immune system and strengthen the gut barrier19.
2'-FL seems to be one of the best HMOs at supporting gut barrier protection20. Following a challenge, 2'-FL increased mucin production in goblet cells, which are known to strengthen the intestinal barrier18.
In addition, it selects for growth of Bifidobacterium strains; these in turn ferment the HMOs and produce metabolites such as short-chain fatty acids. These metabolites can further contribute to strengthening the intestinal barrier21.
MATURES THE IMMUNE SYSTEM
HMOs interact with immune cells both locally in the intestine and systemically, supporting immune cell balance and immune signaling pathways. Their effects are mediated both via direct interactions with immune cells and via metabolites generated during HMO fermentation.
Clinical publications have reported 2'-FL to promote respiratory comfort in infants22, 23. It’s immune-related mechanisms are likely multifactorial. It modulates immune responses, for example by influencing TLR-3 and TLR-4 receptors as well as various T cells19. And it can reduce the number of immune modulating cytokines23. 2'-FL has been found systemically, both in plasma and urine23.
In addition, 2'-FL may support immune function indirectly by promoting the formation of immunomodulatory microbial metabolites24.
SUPPORTS BRAIN DEVELOPMENT AND COGNITION
HMOs are a source of sialic acid and fucose, both of which are essential components for brain development. They also play key roles in neurological processes. A small proportion of certain HMOs can enter the bloodstream, where they may exert positive systemic effects. In addition, HMOs are fermented into microbial metabolites that influence the microbiota–gut–brain axis.
As mentioned earlier, 2'-FL can enter systemic circulation, suggesting that it exerts systemic effects25. It’s been shown to be linked with roles in brain development26. Early exposure to 2'-FL has been positively associated with motor skill development scores27. Higher levels of 2'-FL in early life are also associated with improved cognitive development scores in infants28, 29.
PERFORMANCE IN ADULTS
2'-FL has been investigated in multiple adult clinical studies. The available data demonstrate beneficial effects across several physiological systems2. It shows clear prebiotic activity by increasing beneficial bacteria and thereby promoting the production of favorable microbial metabolites, such as SCFAs. SFCAs have been detected in both stool and blood samples30, 31.
Clinical studies report improvements in gastrointestinal quality of life32, mood33, weight management34and weight management, and supporting healthy insulin and HDL-cholesterol32.

More about HMO science
Breastmilk is the gold standard in infant nutrition. A major difference between milk from humans and other mammals is the amount and abundance of oligosaccharides. Back in 2015, when we were the first to make HMOs commercially available with our MyOli brand®, HMOs were mainly known for their prebiotic benefits.

Breast milk nourishes baby’s microbiome
Mother’s milk has evolved to give a baby and its microbiome a good start in life. Human milk oligosaccharides can’t be digested by the body— only by bacteria in microbiome.
HMOs comprise about 10% of milk solids and are the third largest solid component.

Over 200 HMOs belong mainly to three classes
The vast majority of HMOs belong to one of three classes. 65-70% contain fucose and are called Fucosylated HMOs. About 30% are split between Sialylated and neutral, non-fucosylated HMOs. SL HMOs contain sialic acid. Neutral, non-fucosylated HMOs have neither fucose or sialic acid, and are building blocks for more complex molecules5.

Three HMO classes with many health benefits
Each HMO is unique and offers multiple benefits. In general, 2FL HMOs tend to be good at protecting against unwanted microbes. The sialic acid in SL HMOs is important to brain and cognitive function. Neutral HMOs nurture a healthy microbiome35. This is just the tip of the iceberg for HMO health benefits.

Leading the science
As much as is known about HMO health benefits, more is waiting to be discovered. Some of our own scientists and collaborative partners are world-renowned for their HMO research. We also have a donation program so independent researchers can conduct their own studies.
