3'-SL is a sialylated HMO thought to support early brain tissue organization1 and early life language development2. Research shows it also offers benefits for people beyond early life, for example, in supporting healthy cholesterol and the immune system. These benefits come from its direct bioactive ability; the structure of the molecule itself is the cause of the benefit instead of it being consumed by bacteria which then produce the benefit3.

3'-SL is one of the most abundant HMOs in mothers’ milk. Contrary to most HMOs, its abundance remains consistent independent of time4.

ABOUT HMOs: The third largest component of mothers’ milk solids, HMOs are unlike regular sugars and don’t provide energy to the infant. Rather, they are prebiotics that feed an infant’s microbiota. They are also bioactive and deliver direct health benefits on their own. Every mother’s milk has a unique combination of HMOs, dependent on genetics, nutrition, lactational stage, mother’s lifestyle, geography and more5.

Health benefits

GUARDS AGAINST UNWANTED MICROBES
HMOs have multiple methods to rid the body of unwanted microorganisms, such as acting as decoys or altering epithelial cell gene expression6, 7. Each HMO has a highly specific mode of action8.

3'-SL acts as a strong soluble receptor decoy to block attachment to unwanted microbes.

ESTABLISHES AN OPTIMAL GUT MICROBIOME

HMOs are prebiotics that promote beneficial bacteria growth9, 10. Long known to attract the healthy Bifidobacteria11, 12, recent studies highlight that over 100 non-Bifidobacterium species also use HMOs for their growth13. These bacteria then thrive and produce the metabolites necessary to support good health14.

Compared to many other HMOs, 3'-SL promotes only moderate growth of some bacteria like infant-type Bifidobacteria B. infantis and B. bifidum15, 16. This suggests that 3'-SL’s primary benefit is something other than as a prebiotic. For example, a study suggests it may help stabilize commensal microbial communities under stress conditions and modulate behavioral stress responses via the microbiota-gut-brain axis17.

PROMOTES A HEALTHY GUT
HMOs may promote mucus production18, thereby supporting the physical barrier between the intestinal epithelium and gut content. As well, metabolites produced by HMO fermentation, like short-chain fatty acids, can modulate the immune system and strengthen the gut barrier19.

It’s thought that 3'-SL modulates the gut barrier, possibly by influencing tight junction proteins and mucin production20.

MATURES THE IMMUNE SYSTEM
HMOs interact with immune cells both locally in the intestine and systemically, supporting immune cell balance and immune signaling pathways. Their effects are mediated both via direct interactions with immune cells and via metabolites generated during HMO fermentation.

3'-SL is associated with improving persistent, mild immune troubles21. Like 6'-SL, it also helps shape immune activity, for example, by promoting TH1 and TH17 responses, both of which stimulate the immune system to fight. Both 3'-SL and 6'-SL also can reduce the TH2 response, which is associated with skin discomfort22.

3'-SL is acting immunomodulatory and may be supportive in cases of low-grade inflammation.

SUPPORTS BRAIN DEVELOPMENT AND COGNITION
HMOs are a source of sialic acid and fucose, both of which are essential components for brain development. They also play key roles in neurological processes. A small proportion of certain HMOs can enter the bloodstream, where they may exert positive systemic effects. In addition, HMOs are fermented into microbial metabolites that influence the microbiota–gut–brain axis.

3'-SL may play an important role in brain development. It’s been consistently associated with supportive effects on brain structure. For example, 3'-SL has been linked to maturation of cortical gray matter and developing white matter23. In addition, higher levels of 3'-SL in early life are positively associated with language development24, indicating that it also influences cognitive outcomes.

PERFORMANCE IN ADULTS
In an adult clinical study, 3'-SL was evaluated for its effects on bone health. At a daily intake of 0.6 g, it was shown to improve knee comfort during movement and improve overall mobility25.

In addition, the safety and tolerability of HMOs have been repeatedly confirmed in clinical studies.

See what science tells us

HMOs represent the beginning of complex processes where it's difficult to determine cause and effect. Much more research is needed to clarify their individual and collective benefits. Here’s a summary of what we know now.

MyOli® 3-SL is part of our MyOli® 5 HMO Mix, which is custom blended for your specific target market.

More about HMO science

Breastmilk is the gold standard in infant nutrition. A major difference between milk from humans and other mammals is the amount and abundance of oligosaccharides. Back in 2015, when we were the first to make HMOs commercially available with our MyOli brand®, HMOs were mainly known for their prebiotic benefits.

  • HMOs are the third largest component of breast milk

    Breast milk nourishes baby’s microbiome

Mother’s milk has evolved to give a baby and its microbiome a good start in life. Human milk oligosaccharides can’t be digested by the body— only by bacteria in microbiome.

HMOs comprise about 10% of milk solids and are the third largest solid component.

  • The three HMO classes

Over 200 HMOs belong mainly to three classes

The vast majority of HMOs belong to one of three classes. 65-70% contain fucose and are called Fucosylated HMOs. About 30% are split between Sialylated and neutral, non-fucosylated HMOs. SL HMOs contain sialic acid. Neutral, non-fucosylated HMOs have neither fucose or sialic acid, and are building blocks for more complex molecules9.

  • Health effects of HMOs

Three HMO classes with many health benefits

Each HMO is unique and offers multiple benefits. In general, FL HMOs tend to be good at protecting against unwanted microbes. The sialic acid in SL HMOs is important to brain and cognitive function. Neutral HMOs nurture a healthy microbiome26. This is just the tip of the iceberg for HMO health benefits.
